This protocol is adapted from "Ex vivo Ca2+ 2PLSM measurements with genetically encoded probes" ([dx.doi.org/10.17504/protocols.io.5jyl8pby7g2w/v1](https://dx.doi.org/10.17504/protocols.io.5jyl8pby7g2w/v1)) and modified for imaging genetically encoded sensors (e.g. GRABACh3.0) in ex vivo brain slices with two-photon laser scanning microscopy (2PLSM).
