Loss of dopamine neurons in Parkinson's disease is linked to mutations in *LRRK2* and *GBA1*. Both mutations inhibit Hedgehog signaling, reducing production of neuroprotective factors. This reveals a common mechanism contributing to PD pathogenesis.
