Cryo-EM structure of soluble VPS13C suggests its regulation by a conformational switch and by calmodulin

VPS13C, whose dysfunction caused early onset Parkinson's, is a bridge-like lipid transfer protein with a role in repairing damaged lysosomal membranes. It comprises a bridge-like lipid transfer domain as well as regulatory modules. Here we used in vitro and structural biology techniques to visualize intact VPS13C, obtaining insights as to how these modules regulate VPS13C function.
